Population-scale off-target assessment of Cas9 and other gene editing enzymes 

In addition to creating desired edits at targeted genomic loci, gene editors can also exhibit activity at unintended, off-target loci.  Therefore, off-target risk assessment is a key component of both experimental and therapeutic gene editing strategies.  In this talk, I will describe a method, OligoNucleotide Enrichment and sequencing (ONE-seq), that can identify potential genomic off-target sites of Cas9, Cas12, and base editors while accounting for population-scale genetic diversity.
